GS-441524 is a chemically synthesized nucleoside analog with specific inhibitory effects on feline coronavirus (FCoV). Its antiviral action is targeted at the RNA-dependent RNA polymerase, a key enzyme required for FCoV to replicate in cat cells. This enzyme is responsible for catalyzing the synthesis of viral RNA from the viral RNA template, which is an essential step for FCoV to complete its replication and proliferation. GS-441524 acts as an alternative substrate for this viral enzyme; after being incorporated into the newly synthesized viral RNA chain, it terminates the chain extension due to its structural characteristics, thus effectively blocking the replication process of FCoV and reducing the viral load in the cat's body.

As the main active metabolite of the antiviral drug remdesivir, GS-441524 has a half-life of 24 hours in the mammalian body, which means that the drug can maintain an effective antiviral concentration in the cat's body for a long time after a single administration, which is conducive to the continuous inhibition of viral replication. In the treatment of feline infectious peritonitis (FIP), GS-441524 can directly act on the FCoV-infected mononuclear phagocytes, inhibit the viral replication in the cells, prevent the virus from spreading to other tissues and organs, and alleviate the inflammatory response and tissue damage caused by viral infection, so that the cat's immune system can gradually restore its normal antiviral function and clear the virus in the body.

About Feline Infectious Peritonitis (FIP)


Feline Infectious Peritonitis (FIP) is a severe and fatal systemic viral infectious disease in cats, caused by the mutation of feline coronavirus (FCoV). FCoV is a common virus in cat populations, and most cats are infected with FCoV in their life, but only a small number of infected cats will develop FIP. The key factor leading to the occurrence of FIP is the defect or suppression of the cat's immune function. When the cat's immune system is unable to effectively control the replication of FCoV, the virus will mutate in the body, and the mutated virus will have the ability to invade the systemic mononuclear phagocyte system, leading to the occurrence of FIP.

FCoV is mainly transmitted through the fecal-oral route, and the transmission is more frequent in multi-cat environments such as cat shelters, breeding catteries, and pet shops. The virus can survive in the environment for a certain period of time, and cats can be infected by contacting contaminated cat litter, food bowls, and other utensils. According to the clinical manifestations, FIP is divided into effusive (wet) and non-effusive (dry) types. The effusive type is characterized by the accumulation of exudate in the abdominal or thoracic cavity, which is the most common clinical type of FIP; the dry type is characterized by the formation of granulomas in multiple organs, and the clinical symptoms are more complex and diverse, often accompanied by ocular and neurological symptoms.

Application and Dosage of GS-441524
